In-vivo SPF testing means the SPF number was measured on real human skin, not estimated by an instrument reading a plastic plate. A credible SPF claim rests on human substantiation, and where a brand works to that standard it shows up in the small print: an SPF claim tied to a named test method, UVA marked properly, water resistance in minutes instead of words like "waterproof" or "all-day". Here is what each one means, plus a five-minute check for any bottle before it goes in the trolley.

What does "in-vivo SPF testing" involve?
Picture it. A panel of adult volunteers, small patches marked on their backs. One set of patches gets a measured film of sunscreen — 2 milligrams per square centimetre, spread evenly. The control patches get nothing. A solar simulator delivers graded doses of UV. Then everyone waits. Sixteen to twenty-four hours later, a trained assessor reads which patch turned faintly pink first.
The smallest dose that produces visible redness is the minimal erythemal dose. Divide the protected MED by the unprotected MED and you have the SPF. It is a biological measurement of the one thing you care about — how much longer that skin took to burn.
The internationally used method, ISO 24444, needs at least ten valid subjects, and the result is reported with a statistical confidence interval. No formula scrapes through on one flattering volunteer. All of this takes weeks and costs real money, which is why some SPF numbers in the market have historically rested on cheaper in-vitro screening instead.
In-vivo vs in-vitro: what each one can honestly tell you
I use in-vitro testing constantly during formulation, and I'll defend it in that job. When you're screening twelve prototypes to see which UV filter grid and which film-former hold together, you don't put twelve prototypes on human backs. You spread them on a roughened PMMA plate and read UV transmission. Fast, cheap, directionally useful.
What it isn't is a labelling claim. Plate roughness. How the film settles. The pre-irradiation step. The spreading hand of whoever is on the bench that morning. Each one nudges the number, and two competent labs can hand you two different in-vitro SPFs for the same sample. Human skin is messier than a plate. It is also the surface the product is sold for.

What to look for on your child's sunscreen label
Sunscreens in India are regulated as cosmetics, and BIS standards sit underneath those rules — check the current BIS text for exactly what applies on any given date. Where a brand substantiates in-vivo rather than settling for a screening number, four things look different on the pack.
1. The SPF number gets a source. "SPF tested in-vivo". An ISO method number. Or plainly, "tested on human volunteers". A number floating on its own, with nothing behind it, starts to stand out.
2. UVA gets marked separately. SPF describes UVB only — the burning wavelengths. UVA is the one that drives long-term damage, and it comes through window glass, which matters on a school bus at noon. Look for a PA rating (PA+ through PA++++) or a stated broad-spectrum method. The widely used benchmark is UVA protection of at least one-third of the labelled SPF.
3. Water resistance becomes a number, not an adjective. Tested properly it is reported in minutes — typically 40 or 80 — by re-running the SPF test after timed immersion in water. "Waterproof" and "sweatproof" aren't defensible claims for any sunscreen on earth.
4. Very high SPF claims get harder. Once a number has to survive a human panel, the arms race cools off. Most labels worldwide stop at 50+, because in real use the difference between 50 and 100 is swamped by how thinly people apply.
The five-minute label check to run before you buy
Do this standing in the aisle. It takes about as long as reading a WhatsApp forward, and it filters out most of the noise.
- Flip the bottle first. The front of a pack is marketing. The back is the regulated part, and everything below lives there or on the carton.
- Find how the SPF was tested. "In-vivo", "tested on human volunteers", or a named ISO method. If the pack says nothing, note it and carry on down the list — you'll ask the brand directly.
- Check the UVA marking. A PA rating or a broad-spectrum statement. For Indian daylight, PA+++ is the floor I'd want on a child.
- Read the water-resistance line. 40 minutes or 80 minutes, stated. "Waterproof" tells you something about the rigour of the whole label.
- Read the actives. For a mineral sunscreen you want zinc oxide and/or titanium dioxide named, with their percentages. Under three, mineral filters are the simpler, better-tolerated choice.
- Check who made it. A manufacturer name, an Indian address, a manufacturing licence number. A brand that owns its factory can answer a question about a batch. A brand that doesn't, often can't.
- Check the date of manufacture and shelf life. A half-used bottle from two summers ago is not the product that was tested.
The three questions to ask any brand
Email them, or WhatsApp them. If the data exists, a straight answer takes a minute: Was the SPF determined in-vivo, and by which method? How is UVA protection substantiated? What is the water-resistance duration in minutes? A vague reply tells you as much as a lab report would.
What in-vivo testing still can't do for you
Here is the limit of the whole exercise. That 2 mg/cm² film in the lab is thick — you'd notice it sitting on your own arm. Most of us apply between a quarter and half of it. Protection doesn't fall in a straight line with thickness, so a beautifully in-vivo-tested SPF 50, rubbed on in a rushed smear at the door, behaves nothing like 50.
The testing method fixes the honesty of the number. The dose is still your job. For a school-age child: roughly two full finger-lengths for face, neck and ears, and about a teaspoon for both arms. Fifteen minutes before you leave. Reapply after the 11am games period, and after any swim — not at 4pm, once the day has already done its work.
UVA also comes through car and bus windows, and through a cloudy monsoon sky. A child in the window seat on a long school commute is taking a dose that a burn-free afternoon hides completely. That is why the PA marking deserves as much of your attention as the SPF.
When to see a doctor
Book a paediatrician or dermatologist if your child develops blistering after sun exposure, a burn covering a large area, fever or vomiting alongside sunburn, or a rash that keeps returning after every sunscreen application. Get advice before using sunscreen on a baby under six months, on broken or eczema-flared skin, or if your child takes any medication that can increase sun sensitivity. A mole or spot that changes shape, colour or size deserves a look too.
